Luciferase activity of transfected cells.....

You can hook up the gene from fireflies that makes the glow (luciferase) to a promoter for whatever gene you are interested in and put this into cells. Then when you try to "turn on" your gene of interest, it turns on the production of the luciferase. You light up the luciferase by adding a chemical and measure how much it glows. The more it glows, the more you (would) have turned on your gene.

Does that work with BE as well as Gene?

Not sure. I'm dealing with a GUS (Glutening of Unknown Source). Gus is an angry cus and he doesn't want to tell me where the gluten came from. :lol::blink::(

Suspecting gluten-free Rice Krispies somehow got me but I don't want it to be true..... :unsure:

P.S. Tony wanted to quit his job after his first night. He and Clark had words but Tony apologized this AM.He was under the (wrong) assumption that Brown got all the $ he earns because every student has to make $2,500 a summer but the students get to use it for incidentals. He's upset that he's working all night shifts but we told him that's life. Tony was also just diagnosed with ANA so we're all going to our PCP with him on Monday. That is probably why his joints always hurt and he's always tired. He unfortunately seems to have my immune system. unsure.gif

Just helped SIL move. She is getting divorced. It really sad because she really has nothing but it took 2 small Uhaul, mini van, hatchback & pick up truck trips to move it all. She moved things because They were "hers" in the divorce, even if she didn't have room for them or a need. She has 25 smallish boxes of books but no bed. She bought 4 big book shelves for the books. :blink:

The funniest part was 7 guys trying to move an old upright piano up 1 skinny flight of stairs. They eventually turned it on its side & got it and one of the guys, trapped in the old elevator. Eventually, it made it to the apartment.

There is some sort of moral about if you have enough testosterone envolved......

BE and I talk about his ex and the stuff she got in the divorce and took because it's "hers". He said she sits in her small condo packed full of "her" furniture and does nothing. Seems to me that if you aren't careful you end up winning a lot of stuff that does nothing but hold unpleasant memories.

It was more sad than what is "hers". Its hard to explain. She has 100's of books that most of us would have given away or sold at a garage sale or passed to a friend. She has a couch that has been in some sort of storage (unit, garage, basement, garage again) for 15 years - Yuck. She doesn't need it because she has 2 wood rockers & 2 pleather chairs.

I felt bad leaving her with this huge stack of boxes that you can't get around. I at least put the clothes in the bedroom, Xmas decor & sewing stuff in the storage closet, kitchen stuff next to the kitchen. Another friend & I offered to come back this evening, crack open one of the bottles of wine she moved & set up the bookshelves & put shelf paper in the kitchen cabinets (clean but yucky). I lied & told her I had a bunch of it I wanted to get rid of since she is more than broke. I was just going to get more. It would be an overwhelming mess in a happy situaton... :(

Sorry. Not funny! J & I have already vented to each other enough. He had brought tools so he could put things together & hang pictures if she wanted. She did seem happy to have him there. She was one of his favorite babysitters when he was a toddler.

I do understand, having been through two divorces. It's a combination of wanting to keep everything that would make the other person happy, and feeling like you've lost so much already you can't let go of the things you have, whether they should be part of your life or not.

Do what you're doing. Let her know that she hasn't lost friends along with this huge chunk of her life. Once she begins to realize that she really is ok, and all still there, she'll be able to let go of all the stuff that she's keeping for protection. Divorce isn't about losing a spouse, it's about losing all the dreams and plans you had. About losing what you thought your life was, who you thought you were. All she has right now for protection are the memories caught up in her stuff. Help her learn to make new memories and she'll be able to see the things as things again.

True, not phunny, but symbolic of the strength we have. One of the reasons I love all you guys is the sheer personal strength you exude. :)

      Interesting I read that toasted kasha groats have nutty flavor which I thought like oatmeal with banana and yogurt. Yes quinoa I have for dinner looking to switch oatmeal to buckwheat for breakfast. I have to look into amaranth

      I've never tried bananas or yogurt with kasha. It would probably work but in my mind I think of kasha as being on the savory side so I always add butter, peanut butter, or shredded cheddar cheese. Next time I make it I will try yogurt and banana to see for myself. Amaranth has a touch of sweet and I like to pair it with fruit. Quinoa is more neutral. I eat it plain, like rice, with chicken stock or other savory things, or with coconut milk. Since coconut milk works, I would think yogurt would work (with the quinoa). I went to the link you posted. I really don't know why they rinse the kasha. I've eaten it for decades and never rinsed it. Other than that, her recipe seems fine (that is, add the buckwheat with the water, rather than wait until the water is boiling). She does say something that I forgot: you want to get roasted/toasted buckwheat or you will need to toast it yourself. I've never tried buckwheat flakes. One potential issue with flakes is that there are more processing steps and as a rule of thumb, every processing step is another opportunity for cross-contamination. I have tried something that was a finer grind of the buckwheat than the whole/coarse and I didn't like it as much. But, maybe that was simply because it wasn't "normal" to me, I don't know.

      The basic seems more like oatmeal. You can also add yogurt banana to it like oatmeal right. What about buckwheat flake cereal or is this better to go with.

The word “transient” usually means it changed or disappeared, which can happen with some inflammatory, seizure-related, migraine-related, vascular, or imaging-artifact situations.  Hopefully they will find nothing serious.
